Female mice carrying the MMTV - PyMT transgene that are either Postn + / + or Postn − / − will be examined for changes in
primary tumor size and the number of spontaneously formed pulmonary macrometastases, which is a replication of the experiment reported in Figures 3A, 3B, and Supplemental Figure 13.
Primary tumor size was measured by weight, and the number of metastases was determined by counting lung metastatic lesions.

Patients were stratified by number of brain metastases (1 vs. 2 - 3),
primary cancer type (melanoma vs. other histology) and pre-operative
tumor size (less than vs. greater than three centimeters).
2) The two repeated experiments analyze the source of POSTN expression in the lung and whether it affects the number /
size of
primary and secondary
tumor formation in a spontaneous mouse model of breast cancer (MMTV - PyMT).
The
primary analysis focused on those who underwent SABR or thoracoscopic sublobar resection (wedge or segmental resections) for a
tumor sized ≤ 2 cm.
Approximately one million cells of each clone were inserted into 0.9 mm capillary tubes and centrifuged to form a small pellet; this amount corresponds to a small
primary tumor, or average
size metastasis.
Scintigraphy can determine both the
size of the
primary tumor as well as the location of the metastases that occur in 65 to 90 % of these dogs.
