cell differentiation have shown that in the presence of human MSC,
Th1 cell secretion of IFN - γ dropped by 50 % compared to cultures without MSC.

We first examined the effect of VPA on
Th1 and
Th17
cell differentiation.
In our study, we observed that VPA treatment inhibited the
differentiation of
Th1 and
Th17
cells.
As shown in Fig. 4A, VPA inhibited the in vitro
differentiation of
Th1 cells.
Therefore, our results suggest a direct suppressive effect of VPA on the
differentiation and immune function of
Th1 and
Th17
cells.
The
differentiation of
Th1 cells was analyzed by intracellular staining of IFN - γ (left panels).
Yu et al. (46) used mice deficient for both T - bet and RORγt as T
cell donors and showed that blockade of both
Th1 and
Th17
differentiation is required to prevent acute GVHD.
Moreover, annexin V / propidium iodide staining showed that VPA only marginally decreased the viability of in vitro — differentiated
Th1 and
Th17
cells at a relatively high concentration (1 mM)(Supplemental Fig. 4A, 4B), whereas VPA readily inhibited the

cells at a low concentration (0.2 — 0.5 mM), indicating that apoptosis induction was not the primary mechanism for
