Our model should allow studies of the impact of
aneuploidy on molecular adaptations and cellular fitness.IMPORTANCE Aneuploidy is usually detrimental in multicellular organisms, but in several microorganisms, it can be tolerated and even beneficial.
Using Plk4 overexpression (PLK4OE) during epidermal development, we assess the impact of centrosome amplification and
aneuploidy on skin development and tumorigenesis.
The researchers also investigated the impact of
aneuploidy on other biological pathways, such as transcription, the first stage of gene expression in which a segment of DNA is copied into RNA.

Depending
on the genes that are missing or duplicated, some people can survive with
aneuploidy.
However, even when survival is possible,
aneuploidy can have a significant impact
on a person's intelligence, health and lifespan.
Several researchers interviewed, after asking to go off the record, erupted in venomous attacks
on Duesberg, saying that
aneuploidy is the result of cancer, not the cause.
In 2005 Duesberg was invited by the National Institutes of Health — which had long since dismissed him and his research — to give a grand rounds presentation
on his
aneuploidy work.
The cells» response to environmental changes also varied considerably, suggesting that
aneuploidy has an impact
on the robustness of many, if not all, biological processes.
«The organization of cell colonies and phenotypic switching between different types of colonies becomes a lot more flexible and rapid with reversible
aneuploidy than if it depended
on random mutations in the genes,» Skupin says.
Partly based
on his MIT Ph.D. thesis research performed in yeast, Sheltzer suspects
aneuploidy causes errors in DNA replication, as well as problems with chromosome segregation during cell division.
In contrast to the view that
aneuploidy is deleterious, the aneuploid unisexually generated isolates MN35 (n +113) and MN55 (n +19) were as virulent as the euploid wild - type parent, despite their modest TS growth at 37 °C
on YPD rich media in vitro (Figure 7).
My post-doctoral work
on the identification of genes required for normal germ line development and fertility led to the discovery that the germ line is exquisitely sensitive to mutations in components of the mitotic spindle that have the potential to lead to
aneuploidy — this sensitivity may also extend to embryonic and adult stem cells.
«The authors went
on to study the resulting blastocysts for
aneuploidy (frequency of abnormal chromosomes), and pattern of gene expression (which genes are active and at what level).
The team's work highlights this by presenting a comprehensive view of genome evolution
on many different levels (e.g., differences in ploidy,
aneuploidy, genetic variants, hybridization, and introgressions) that is difficult to obtain at the same scale and accuracy for other eukaryotic organisms.

The alternation of two life stages in different environments (extracellular promastigotes and intracellular amastigotes) offers a unique opportunity to study the impact of environment
on aneuploidy and gene expression.
The method provides quantitative
aneuploidy calls, and is integrated into an R software package available
on GitHub and as an Additional file of this manuscript.
