More recently, as scientists realized that IDE was also involved in
clearance of amyloid - beta, they have begun searching for ways to supercharge the enzyme to see if it could prevent the build - up of the amyloid plaques that are a hallmark of Alzheimer's disease.
This year researchers finally demonstrated
clearance of amyloid - β in humans, after a long series of failures.
CONCLUSION: In this Lewy body variant case, with globally stable functional and cognitive features, Abeta immunization resulted in a significant
clearance of amyloid deposits, with remaining tau and synuclein pathological features in the brain.
Regardless, simply through the demonstrated
clearance of amyloid in human patients this is a big step forward for the field.
Previously, other researchers reported that ApoE4 seemed to reduce
the clearance of amyloid - β, the protein that clumps together to form the hallmark plaques of AD, from the brain in a mouse model.
One 2013 study showed that the extracellular space in a mouse's brain expands by 60 percent during sleep, and
clearance of amyloid plaque (one protein implicated in Alzheimer's) spikes.

Compared with control groups, the mice that were given oleocanthal showed significantly enhanced
clearance and degradation
of the beta -
amyloid peptides.
The researchers conducted a series
of experiments to see if they could replicate the phenomenon
of amyloid beta
clearance absent brain inflammation.
«In the sporadic form
of the disease, we think the problem isn't necessarily with the generation
of amyloid - beta, but possibly with its
clearance.»
These new findings shift the emphasis to
clearance of beta -
amyloid.
In the new study, they measured beta -
amyloid production and
clearance rates in 12 patients with late - onset Alzheimer's and 12 age - matched volunteers free
of the disease.
However, the
clearance of beta -
amyloid was about 30 % slower in those with Alzheimer's disease than in cognitively normal people.
SIRT1, in turn, was thought to reduce the accumulation
of amyloid - β by activating another protein — a possible mechanism behind the reduced
amyloid - β
clearance observed in AD.
«This study is significant in that it reports the first measurement
of beta -
amyloid production and
clearance in Alzheimer's,» says Dr. Marcelle Morrison - Bogorad, director
of the Division
of Neuroscience at NIH's National Institute on Aging (NIA).
Other theories and lines
of research have begun to prosper due to the lack
of tangible human results for anti-
amyloid immunotherapies, in particular that neurofibrillary tangles
of misfolded tau protein are just as much a target for
clearance as is
amyloid - β, and that perhaps it is time to focus on the decline
of known
clearance mechanisms rather than the
amyloid itself.
Still, over the past couple
of years, one
of the latest entries to the class
of anti-
amyloid immunotherapies has lived up to some
of the promise first seen in animal studies
of amyloid clearance.
However, the new inhibitory neurons did not affect
amyloid beta levels, suggesting that the cognitive enhancement did not occur as a result
of amyloid clearance, and
amyloid did not impair the integration
of the transplant.

The study suggests that «a slow - gamma deficit might be part
of this [Alzheimer's disease] pathogenesis [and that] manipulating slow - gamma activity... could be a new way to suppress
amyloid - β production and increase
amyloid - β
clearance,» Huang added.
We can hope that as the first effective therapies make it into the clinic, most likely for the
clearance of forms
of amyloid, there will be a growing enthusiasm for work on ways to remove other types
of metabolic waste.
Researchers have shown that
clearance of the substance
amyloid - beta that forms plaques is impaired in individuals with Alzheimer's.
There was a significant difference
of the average
clearance rate for
amyloid - beta between the 12 early Alzheimer's individuals and the 12 normal individuals.
Amyloid - beta was produced at the same average rate by both groups, but in the Alzheimer's group there was an average drop in the
clearance rates
of about 30 %.
Rats undergoing natural sleep or sleep induced by anesthesia increased their brain's interstitial spaces by 60 %.6 This leads to significant exchanges between CSF and interstitial fluid that result in increased rates
of beta -
amyloid clearance during sleep.
The peripheral sink Aβ hypothesis indicates that the peripheral
clearance of Aβ and its regulation by dietary phytosterols is
of substantial interest since it may delay hypercholesterolemia and the early onset
of amyloid plaque development.
