Finally, we are undertaking a vast mutational analysis of
endocytic proteins in cancer, to define genetic alterations affecting the endocytic machinery.
Using automated high - content superresolution imaging and quantitative data analysis, we determined how more than a dozen
endocytic proteins are organised at the nanoscale.

However, approximately 40 % of human (and most eukaryotic)
proteins localize to chemically distinct subcellular environments, including the organelles that compose the secretory pathway,
endocytic vesicles, mitochondria, lysosomes or they are secreted into the extracellular milieu.
We thus propose a molecular hypothesis — termed the GL - Lect hypothesis — on how GlycoLipids and Lectins drive the formation of compositional nanoenvrionments from which the
endocytic uptake of glycosylated cargo
proteins is operated via clathrin - independent carriers.
We hypothesized that certain lectins link glycosylated cargo
proteins and glycosphingolipids into molecular nanoenvironments from which
endocytic pits are formed without the help of the cytosolic clathrin machinery (Figure 1).
In the
Endocytic Trafficking and Intracellular Delivery team we study the cell biology of carbohydrate - binding
proteins (lectins) at the level of cellular entry though clathrin - independent endocytosis, retrograde sorting on early endosomes, and translocation to the cytosol.
Our group has discovered two
protein -
protein interaction networks that sit at the heart of numerous
endocytic and trafficking processes: the EH (Eps15 Homology) and monoUb networks (9 - 13).
