The researchers hoped to create a cellular model to parse the effects of specific gene mutations, according to a press release, but in the process they discovered that skin cells
from FOP patients were much more readily converted into iPSCs than cells from healthy individuals.
To confirm their hunch, the researchers prevented BMP signaling, which resulted in fewer iPSCs being generated
from FOP patients» cells.
Surprisingly, the scientists discovered that they could create more iPSCs from cells taken
from FOP patients than those taken from healthy individuals.
However, chondrogenesis assays in chick micromass cultures showed that enhanced signaling by the mutant R206H ACVR1 can be partially inhibited by the BMP antagonist Noggin, suggesting that in addition to BMP - independent activity by the R206H ACVR1 receptor, this mutant receptor is also responsive to BMP, consistent with previous BMP signaling pathway studies that used cells derived
from FOP patients (48 — 50).
He is now trying to verify the findings in cells
from FOP patients and healthy people.
Unlike the skin cells from which they originated, the human iPS cells created
from FOP patients show increased cartilage formation and increased bone mineralization, two critical steps that are necessary to form mature bone.
