First, after culturing human peripheral blood mononuclear cells (hu - PBMCs) with the IL - 15 superagonist, flow cytometry revealed increases in
granzyme B and perforin expression in the CD3 -, CD16 +, CD56 + NK cells.
In particular, they investigate how killer immune cells use pore forming protein perforin, antimicrobial peptide granulysin, and immune protease
granzyme B in synergy to destroy pathogens in host cells.
To assess
granzyme B production, CFSE - labeled cells were stimulated for 24 or 72 h, surface - stained as described above, permeabilized using a Cytofix / Cytoperm kit (BD Pharmingen), and then stained with flurochrome - labeled
granzyme B Ab (GB11; Life Technologies) at 4 °C for 30 min prior to flow cytometric analyses.
Deletion of DGKζ or Cbl - b comparably enhanced CD8 + T cell functional responses, such as proliferation, production of IFN - γ, and generation of
granzyme B when compared with wild type T cells.

granzyme B was measured from purified T cells gated from CD8 + populations after 24 and 72 h of stimulation with 0.3 μg / ml anti-CD3 stimulation.
Further, although we could detect enhancement of
granzyme B production in DKO T cells relative to WT, DGKζ − / −, or Cbl - b − / − T cells at early time points (Fig. 3B, upper panels), we did not observe enhanced
granzyme B production in DKO relative to DGKζ − / − or Cbl - b − / − T cells at later time points (Fig. 3B, lower panels), at higher or lower concentrations of anti-CD3 Ab, or with addition of anti-CD28 Ab (Supplemental Fig. 2B).
Consistent with previously reported analyses, we also observed enhanced IFN - γ and
granzyme B production in DGKζ and Cbl - b — deficient T cells.
Perforin pores in the endosomal membrane trigger the release of endocytosed
granzyme B into the cytosol of target cells.
They kill cells by releasing small cytoplasmic granules of proteins called perforin and
granzyme that cause the target cell to die by apoptosis (programmed cell death).
Lieberman, Dotiwala and their team were particularly struck by how much
the granzyme - sparked cascade resembles apoptosis, a controlled form of cellular suicide that helps eliminate damaged or potentially cancerous cells.
Lieberman notes that her laboratory reported a similar reaction to perforin, granulysin and
granzyme in intracellular bacteria in a 2014 Cell paper.
They also found elevated
granzyme A levels in blood samples taken from non-human primates infected with chikungunya, as well as from human chikungunya patients.
Exploring further, the scientists showed that mice missing
the granzyme A gene, when infected with chikungunya virus, experienced dramatically less foot swelling and arthritis.
Overall the findings suggest that
granzyme A could serve as a potential drug target for anti-inflammatory treatments for chikungunya — and perhaps also for other inflammatory diseases.

granzymes, which stimulate apoptosis, or cell death.
