Many, if not most of the genomic loci associated with human traits lie
in noncoding regions.
Undoubtedly a significant proportion of important genetic variants lie
in noncoding regions, where they exert a regulatory effect.
One of the noncoding de novo mutations was 254 kilobases downstream of FGFR2,
in a noncoding region that looks (according to chromatin marks) like a neural crest enhancer.
This, in my opinion, is one of the most compelling parts of this study: in vivo functional validation of a single base change
in a noncoding enhancer that's hundreds of thousands of bases away from the gene it regulates.
But full sequencing will soon be just as cheap as exome sequencing and will catch disease mutations
in noncoding regions as well, Gibbs notes.
At first, Sprecher suspected that either they had performed the genetic analysis incorrectly or the missing mutation was hiding
in a noncoding or «junk» region of the genome.

Sebat says the large
noncoding portion of our DNA — often previously referred to as «junk DNA» — has so far been ignored
in autism research.

The differences between coding versus
noncoding trees were not solely due to shorter sequence length of the coding data, because the full coding data set (13.3 million bp for c123) produced a tree with fully supported (100 % BS) relationships that were incongruent with those fully supported
