«We think that a therapy that would control the number and properties of
myofibroblasts in the heart might be useful, but we don't know that for sure,» Genin adds.
Fibroblasts also convert to
myofibroblasts in response to high blood pressure, or hypertension.

The most common cells found
in healing wounds are
myofibroblasts, which were thought to only form a scar.
The orbital remodeling
in TED is likely induced by infiltrating T lymphocytes and mast cells, which activate orbital fibroblast effector cells to either proliferate and form scar - producing cells called
myofibroblasts or turn into pro-inflammatory fat cells.
A new study published
in The American Journal of Pathology found that activation of the aryl hydrocarbon receptor (AHR) pathway by its ligands blocks collagen production and
myofibroblast proliferation
in TED.
Previous research has demonstrated that Akt1 is at least partly responsible for the development of
myofibroblasts, cells that migrate to the sites of injury to aid
in wound healing.
Rare stem cells (
in red) that give rise to scar - tissue secreting
myofibroblast cells, here found near the blood vessels of a mouse kidney (
in green).
«Previous research indicated that
myofibroblasts are the cells responsible for fibrosis,» said Benjamin Humphreys, MD, PhD, senior author of the research paper and a physician scientist
in the Renal Division at BWH.
The new fibroblasts
in the scar are activated and adopt a smooth muscle actin — and periostin - positive «
myofibroblast» phenotype, which appears to last as long as the scar is not mature (~ 10 days after MI).
The researchers, led by Benjamin Humphreys, MD, PhD, found that a rare population of stem cells located outside of blood vessels
in mice become
myofibroblast cells that secrete proteins that cause scar tissue.
One long - held hypothesis was that the stem cells that give rise to
myofibroblasts are found
in the bone marrow, but Humphreys» research disproves that.
Cardiac
myofibroblasts differentiated
in 3D culture exhibit distinct changes
in collagen I production, processing, and matrix deposition.
Counteracting effect of TRPC1 - associated Ca2 + influx on TNF - α - induced COX -2-dependent prostaglandin E2 production
in human colonic
myofibroblasts.
The researchers found that a specific stem / progenitor cell population was inhibited following radiation
in mice missing Nrf2, and that alveolar type 2 cells
in these mice were more likely to change into
myofibroblasts — a cell type implicated
in fibrosis.
The WUSTL research focuses on the role
in heart disease of role - switching cells called
myofibroblasts that proliferate
in over-stressed or injured hearts.
If blood pressure is high enough to provoke fibroblasts to become
myofibroblasts, the cells also may get stuck
in their helper state.
But heart cells never truly regenerate
in the damaged tissue, and
myofibroblasts compensate for their absence by forming a stiff, collagenous scar that interferes with the heart's ability to maintain stable heart rhythms and to expand and contract forcefully to pump blood.
These cells, which were named
myofibroblasts to reflect their double nature, secrete fibers to fill
in a wound and then contract to bring together its edges.
«Drugs that block the effects of
myofibroblasts on the electrical or mechanical properties of heart tissue or that coax them to revert to fibroblasts might be more effective than current therapies,» says Guy M. Genin, PhD, associate professor of mechanical engineering and materials science
in WUSTL's School of Engineering & Applied Science, who is one of three co-primary investigators (PI) on the grant.
Myofibroblasts can proliferate elsewhere
in the body as well — although they may arise from different cell types
in different tissues — and fibrotic remodeling of the kidney, liver (cirrhosis of the liver) and lungs follows a similar progression, Genin says.
Moreover, although CXCR4 inhibitors significantly reduce
myofibroblast content
in all BC subtypes, they decrease angiogenesis only
in HER2 BC.
Although
myofibroblasts expressing CXCL12 are associated with the presence of axillary metastases
in HER2 breast cancers (BC), the therapeutic interest of targeting CXCR4 / CXCL12 axis
in the different BC subtypes remains unclear.
As a proof of principle, we demonstrated correlation of the high - risk genotype at the statistically most strongly associated variant with decreased secretion of the soluble WNT - antagonist SFRP4,

myofibroblasts.
The effects of therapeutic lasers
in wound healing include vasodilation, angiogenesis, increased collagen synthesis by fibroblast, differentiation of fibroblast into
myofibroblasts, stimulation of leukocytes, and enhanced antioxidant activity.
