Whitehead Institute researchers report that 10 long
noncoding RNAs (lncRNAs) play a vital role in the regulation of white fat cells.

Genome editing technology enables precise modification of individual protein coding genes, as well as
noncoding regulatory sequences, enabling the elucidation of functional effects in human disease relevant cellular systems.

The authors also leveraged our current knowledge of gene - phenotype relationships, and the comprehensive nature of WGS data, to identify three pathogenic
noncoding variants.
The HGP and subsequent research efforts have changed the consensus view of genes and
noncoding DNA, casting them as part of an increasingly complex image of genes, DNA and other components of the genome.
It suggests that all of

It's a complicated picture, to be sure, but it suggests some specific areas in which
noncoding variants are exerting a cis - regulatory effect on the LPA gene.
In addition, the majority of HARs are
noncoding — they do not appear to encode for specific genes.
Sessions will cover recent findings in RNA polymerase II regulation, including the contributions of
noncoding RNAs, chromatin structure and post-translational modifications, and other factors that regulate gene expression.
As our understanding of
the noncoding portion of the genome improves, it will become even more apparent that whole - genome sequencing (and not exome sequencing) will be required to characterize the full extent of phenotypically - relevant genetic variation in humans.
We know, of course, that
noncoding portions of the human genome contain functional elements contributing to phenotypic variation.
These included tier 1 (coding), tier 2 (conserved / regulatory), and tier 3 (unique
noncoding) variants.
Rather than modify protein structure and function,
these noncoding variants frequently impact regulatory elements and thereby alter the quantitative and spatiotemporal regulation of gene expression.
Eukaryotic cells produce diverse types of protein - coding and
noncoding RNAs, some of which have a direct role in the regulation of gene expression.
It is as if Saccharomyces cerevisiae has found the philosopher's stone but instead of changing lead to gold, it turns
noncoding DNA into a gene that codes for a partially structured protein.
Here at La Jolla Institute, I am excited to continue utilizing
noncoding RNAs as a tool for better understanding other aspects of T cell biology.
In particular, our research focuses on developmental origins of health and disease with an emphasis on environmental exposures and epigenetic mechanisms (DNA methylation and
noncoding RNA).
Undoubtedly a significant proportion of important genetic variants lie in
noncoding regions, where they exert a regulatory effect.
