To study the effect of hypoxia / reoxygenation cycles on breast cancer, we exposed two metastatic human breast cancer cell lines (MDA - MB 231 and BCM2) to cycles
of chronic hypoxia and nutrient deprivation.
Here, we show that a non-adherent, stem - like, and metastatic CSC - enriched subpopulation could be isolated by exposing human metastatic breast cancer cell lines to cycles
of chronic hypoxia followed by reoxygenation.

Published in Nature Medicine on May 1, 2017, the study demonstrates
chronic ischemic
hypoxia (lack
of blood and oxygen) after spinal cord injury and how blood flow plays a key role in the cause and treatment
of motor disorders.
Fat tissue in obese people is believed to exist in a
chronic state
of mild
hypoxia because the small blood vessels are unable to supply sufficient oxygen to fat tissue.
The effects
of chronic prenatal
hypoxia on the fetal and adult offspring cardiovascular system are prevented by maternal treatment with vitamin C during pregnancy.
Therefore, the primary novelty
of the discoveries reported here is that programming by prenatal
chronic hypoxia of cardiac and vascular dysfunction in adulthood follows the induction
of oxidative stress in the fetal heart and vasculature, and that cardiac and endothelial dysfunction in adulthood can both be prevented by maternal treatment with antioxidants during pregnancy.
The aims are to study the adaptive response and damage / repair mechanisms
of the foetus and newborn to a poor pre - and perinatal environment as during severe preeclampsia (asphyxia,
hypoxia, growth restriction),
chronic intrauterine infections and perinatal insults related to
hypoxia and inflammation.
Furthermore, susceptibility
of genomic instability can vary after acute or
chronic exposure to
hypoxia followed by reoxygenation [23].
However,
chronic exposure
of cells to
hypoxia induces disassembly
of the replisome, preventing replication restart after reoxygenation [23].
We showed that a
chronic oxidative stress accelerates aging through accumulation
of HIF (
Hypoxia - Inducible Factor) and modulation
of insulin signaling (Gerald, Cell, 2004; Laurent, Cell Metabolism, 2008).
But veterinarians who believe that brachycephalic dogs suffer more from heart issues suggest that a deficiency
of oxygen over time (
chronic hypoxia) causes blood channels in the lungs to narrow (pulmonary vasoconstriction) which then leads to increased blood pressure within the heart (pulmonary hypertension), a known risk for heart failure.
Canine: Determination and Mapping
of Acute and
Chronic Hypoxia in Canine Tumors D08CA - 077, Marlene L. Hauck, Grant Amount: $ 113,230
Dr. Hauk's research will define the genes associated with
chronic and intermittent
hypoxia to give clinicians a tool — based on the type
of hypoxia — to better tailor therapy for individual patients.
