A powerful new tool for the study of
spatial memory was today described in Nature as a significant improvement over the current gold standard, the Morris
Water Maze.
Here, we examined the extent to which performance in the Morris
water maze — the most frequently used behavioral assay of
spatial learning and memory in rodents — translates to humans.
Following three months of treatment, all mice were analyzed for
spatial memory by the two - day
water maze, disinhibition phenotype by the elevated plus
maze and hippocampal dependent memory by fear conditioning.
Results from this two - day
water maze show that AD mice take considerably longer to find the hidden platform on Day 2 than AD mice treated with J147 for three months (Figure 1A), demonstrating that J147 significantly improved the
spatial navigational memory in aged, transgenic AD mice.
J147 and J147 + donepezil rescued the deficit in
spatial navigational memory caused by scopolamine in the
water maze but donepezil alone failed to rescue the deficit (Figure 4B).
They have
spatial learning and memory defects as assessed in the Morris
water maze and the radial arm
maze, show developmental delay in sensorimotor milestones, and exhibit locomotor hyperactivity, lack of behavioral inhibition, and stereotypic behavior.
