Dr. Gnjatic's research focuses on human antigen -
specific immune responses to tumor antigens, in an attempt to define new targets for the development of cancer immunotherapies, assess the efficacy of these immunotherapies, and learn why they may fail.

He also studies the mechanisms of antigen presentation
to T cells, the impact of immunoregulation on
tumor antigen -
specific responses, and characterization of the
tumor -
immune microenvironment.
However, despite this success, the use of immunotherapy remains limited by the scarcity of
tumor -
specific antigens — substances that can trigger an
immune system
response to a particular type of cancer.
Most importantly, many of the T cells were able
to recognize the
tumor cells directly, demonstrating that the vaccine had triggered a
tumor -
specific immune response that could target the patient's
tumor.
«Our results clearly demonstrate a significant, although unexpected, role for
specific gut bacteria in enhancing the
immune system's
response to melanoma and possibly many other
tumor types,» said study director Thomas Gajewski, MD, PhD, professor of medicine and pathology at the University of Chicago.
A personal cancer treatment vaccine that targets distinctive «neoantigens» on
tumor cells has been shown
to stimulate a potent, safe, and highly
specific immune anti-
tumor response in melanoma patients, report scientists from Dana - Farber Cancer Institute and the Broad Institute of MIT and Harvard.
Using transplantable
tumor models expressing antigen in an inducible manner or spontaneous
tumor models expressing nominal antigens, we try
to decipher whether the
tumor is ignored or leads
to deletion, anergy or class switch of the
specific immune response or generate suppressor T cells.
Cancer vaccines are designed
to stimulate an
immune response against
tumor -
specific or
tumor - associated antigens, encouraging the
immune system
to attack cancer cells bearing these antigens.
Through her CLIP Grant, she is analyzing patient samples
to determine how Poly ICLC (a TLR agonist) affects the
tumor environment and
immune responses against
specific mutations,
to identify how Poly ICLC could complement existing immunotherapies.
Therapeutic vaccines are designed
to elicit an
immune response against
tumor -
specific or

immune system
to attack cancer cells bearing these antigens.

Over the past few years, they have extended their research
to understand how products of cellular necrosis and the innate
immune responses they induce affect the
tumor microenvironment, with a
specific focus on ovarian cancer.
There, they will be exposed
to tumor antigens released from dying glioma cells through TK + valacyclovir - induced glioma cell death, and thus mediate a
specific anti-malignant glioma
immune response against remaining malignant glioma cells.
They do, however, play a key role in helping
to boost the anti-
tumor immune response by: (1) expanding the number and improving the function of existing CTLs that are
specific to the same
tumor antigen; and (2) providing help
to other
immune cells, including B lymphocytes that are precursors
to antibodies and natural killer (NK) cells that are also important in the overall anti-
tumor immune response.
We believe DCs are the most important
immune cells
to target because they initiate the
specific immune response that generates CTLs
to kill the
tumor.
