Its interior is lined with a series of gold electrodes, each dotted with thousands of identical short strands of DNA
called aptamers.
Chemistry: Selection of a
DNA aptamer for homocysteine using systematic evolution of ligands by exponential enrichment
Eva Klopf and colleagues demonstrate that RNA polymerase -
binding Aptamers represent a novel type of regulatory RNA signal in Saccharomyces cerevisiae
We have successfully evolved families
of aptamers targeting the HIV envelope that neutralize HIV infectivity, yet can be internalized in HIV infected cells to deliver therapeutic siRNAs.
In fact, the mice that received the complete nanomedicine package had smaller tumors and longer lifespans than their counterparts injected with saline, nanoparticles without the drug, the drug alone, and nanoparticles with the drug but without the
targeting aptamers.
Estrogen molecules caught by the
longer aptamer are likely held above the bilayer, and so do not create the electrical signal.
Finally, the laboratory has been actively using an in vitro evolution process (SELEX) to evolve RNA
aptamers for use as therapeutics but also to serve as siRNA delivery vehicles.
Treating a human disease
with aptamers is still «a leap, but in principle we can do this,» says Smith.
Estrogen molecules that are caught by the
short aptamer disrupt this layer, which in turn changes the current through the device.
And
because aptamers are easily synthesized by machine, Smith says they may be cheaper and easier to make than drugs produced by cell culture.
«We gave huge amounts of
aptamer over six months», yet there were no signs that the compound was unsafe, he says.
The problem seems to be confined to this particular compound, but nonetheless, the company has shifted its focus away
from aptamers.
Their sensor would expose samples to all those
random aptamers, garnering information from each hit.
Szostak's work with
synthetic aptamers and ribozymes has convinced him that RNA could have once dominated the world.
Unlike aptamers, which are capable only of sticking to something else, ribozymes can change the structure of other molecules.
Aptamers turned out to be capable of performing an extraordinary range of tasks.
They say that the system can be constructed with more than five
different aptamers, allowing detection of multiple biomarkers at once.
The team demonstrated that their system can work by testing three
aptamers on one DNA backbone.
If a sample contains gluten, the substance can bind to one of three capture agents, which can be antibodies or a DNA -
based aptamer, that specifically latch onto gluten proteins from certain sources.
Losick is using this year's winning Ph.D. dance — a Scottish folk dance version of directed DNA
aptamer evolution — as a teaching tool in his classes.
Aptamers offer advantages over antibodies as they can be engineered completely in a test tube, are readily produced by chemical synthesis

Our group seeks to investigate the use of chemically - modified nucleic
acid aptamers, single stranded DNA or RNA sequences that specifically bind to a diverse variety of targets, in biosensing and catalysis.
SELEX uses natural selection to find the small strands of DNA
called aptamers (the other dancers) that bind specifically to the target.
Plank and her team attached their estrogen -
binding aptamers to the other important part of their device: the carbon nanotube thin film field effect transistor (CNT FET).
Plank notes that if water was used in the device, instead of the biologically similar buffer, the
long aptamer might also produce a signal because the electrically sensitive layer would be thicker.
This symposium features several topics including extension of the genetic alphabet, selection
of aptamers or DNAzymes, as well as studies in DNA and RNA epigenetics.
Indeed, treatment of mice with POSTN specific
DNA aptamers, single - stranded DNA oligonucleotides designed to bind and inhibit POSTN, was shown to decrease primary tumor growth and metastasis in a xenograft model of mammary tumorigenesis (Lee et al., 2013).
@Vic — Science has repeated evolution, e.g. SELEX / RNA
aptamers, speciation in labs, Lenski's E. coli experiments, artificial selection in domesticated organisms, etc, etc..
The molecule was designed such that the EGFR - targeting component is sandwiched between the HER2 - and HER3 - targeting components in what is known as a HER2
aptamer - EGFR siRNA - HER3 aptamer chimera.
But if a sample is exposed to such
an aptamer library, a complex molecule is going to bind with a lot more sequences than a simple one.
Johnson's team borrowed a technique from cancer biology, called the systematic evolution of ligands by exponential enrichment (SELEX), which creates a huge library of random, short chains of nucleotides, called
aptamers, and then incubates them with a target of choice, such a specific breast cancer cell.
SELEX is typically repeated multiple times, with scientists filtering out
the aptamers that are not specific to their target.
This therapy was
an aptamer, a small strand of nucleotides designed to inhibit TFPI's activity by binding to it, much like an antibody.
But
the aptamers were designed to bind either to an anticancer drug called doxorubicin or an antibiotic called kanamycin.
In it, a continuous stream of a buffer solution flows next to
the aptamer - coated electrodes.
A chemical group at one end anchors
each aptamer to the gold electrode.
When they do,
the aptamers curl around the drug and pull the redox reporter close enough to the electrode for the electrons to hop over and produce a rise in the electric current, signaling the presence of the drug.
Only the drug molecules are small and fleet enough to diffuse through the buffer and get snagged by
the aptamers.
«
Aptamers are a potentially powerful tool for sensors because they are so versatile and selective,» said Natalie Plank, a researcher who studies nanomaterial device fabrication at the Victoria University of Wellington.
The beauty of
the aptamer plus CNT FET sensors is that the aptamers can be easily replaced with new ones that target a different molecule, Plank said.
Aptamers are developed through a process similar to natural selection.
Once the researchers attached
the aptamers to the carbon nanotubes, they tested the devices in a buffer specifically chosen because it has similar properties to biological fluids.
