All co-culture experiments were done in a 1.5
ml microcentrifuge tube with a 20 μl of Schneider's medium with 10 % FBS (Gibco) and 1 % Penicillin - Streptomycin for 4 h at 22 °C.
The kit adopts a novel purification method, using magnetic beads and phosphatidylserine - binding protein to isolate exosomes and other EVs from cell - culture medium and body fluids by a
normal microcentrifuge.
POS were then labelled with AlexaFluor ® 488 Dye (Invitrogen) in 0.1 M sodium bicarbonate / 5 % sucrose in a light -
tight microcentrifuge tube for 1 h at room temperature.
After washing twice with ice - cold PBS, cells were scraped and collected in
a microcentrifuge tube.
